Take care to use the needle valve only to limit inflatable gas amount but never close it.
I've made the experience that you shouldn't open the airpump with closed needle valve
because the airpump's plastic valve wouldn't keep the pressure then and blow it out between the gaskets

Did you have a follow up regarding how you felt after using N2O again?

I'm interested in using an aquarium pump to do a scrotal/genital inflation, but even with a sterile syringe filter, I'm hesitant to add further risks to this process by using ambient air.

I will stay with N2O.

Don't do it. As far as I know, even filtered air stings a lot more than CO2 and needs a lot more absorbtion time.

What's your setup? I'm curious because it's hard to find 16g N2O cartridges. 8gs don't work with the tire inflator as far as I'm aware

Use a bike inflator for unthreaded capsules and normal 8gs for cream dispensers.
They fit perfectly with some little changes:

Needed: A nut for a socket wrench, a coin, a small piece of rolled cardboard

Place the nut with the coin on top into the inflator and the cardboard around

It fits perfectly:

You have to play a little with the size/height of the nut
